Description

Gentalab® Eye and Ear Drops are manufactured by Laborate Pharmaceuticals. The key components of the medication are Gentamicin – an Aminoglycoside antibiotic, derived from an actinomycete, Chlorpheniramine Maleate – a first-generation sedative antihistamine, and Zinc Sulfate – an inorganic compound.

Indications:

- bacterial eye infections (conjunctivitis, keratitis, keratoconjunctivitis, corneal ulcers, blepharitis, blepharoconjunctivitis, acute meibomianitis, and dacryocystitis);

- bacterial ear infections (otitis externa, acute otitis media, chronic suppurative otitis media);

- relieving symptoms of serious allergic reaction called angioneurotic edema, which causes swelling of the eyes.

Gentamicin kills infections caused by Staphylococcus aureus, Staphylococcus epidermidis, Streptococcus pyogenes, Streptococcus pneumoniae, Enterobacter aerogenes, Escherichia coli, Haemophilus influenzae, Klebsiella pneumoniae, Neisseria gonorrhoeae, Pseudomonas aeruginosa, and Serratia marcescens.

Do not use Gentalab® Eye and Ear Drops to treat a viral or fungal infection in the eye or ear.

MOA (mechanism of action):

Gentamicin binds to the bacterial 30S ribosomal subunit. Ribosomes, which produce protein, are composed of two subunits in bacteria, a 30S and a larger 50S. By binding to the ribosome, Gentamicin inhibits the translocation of tRNA during translation and leaves the bacterium unable to synthesize proteins necessary for growth. Gentamicin does not interfere with protein synthesis in human cells. Chlorpheniramine works by preventing the actions of histamine – a substance produced by the body when it reacts to a foreign substance such as pollen or pet fur (an allergen). It blocks histamine receptors and so stops the chain reaction that causes the symptoms of the allergy. Chlorphenamine is called sedative because it causes drowsiness. Zinc Sulfate as an astringent helps reduce redness and irritation.

Dosage

For mild to moderate infections: instill 1 or 2 drops into the treated eye(s) every 4 hours.

For severe infections: instill 2 drops into the treated eye(s) every hour until improvement occurs.

Precautions:

Gentalab® solution is not for injection.

Continue to use Gentalab® Eye and Ear Drops for the full treatment period as advised by your doctor. You should not use Gentalab® Eye and Ear Drops for more than 21 days, unless your doctor says otherwise.

Gentalab® solution is associated with allergic reactions, even following a single dose. Discontinue using drops immediately and contact your physician at the first sign of a rash or allergic reaction.

Prolonged use may result in a fungal infection. If superinfection occurs discontinue use and institute alternative therapy.

When used in the eye, Gentalab® drops may cause your vision to blur temporarily after you have applied them into your eye. Do not drive or operate machinery until this has worn off.

Patients should be well hydrated during treatment.

Avoid contaminating the applicator tip with eye, fingers or another source.

Patients should not wear contact lenses while using Gentalab® solution for eye infection.

Wash your hands before using the eye and ear drops.

Missing a dose

If you miss a dose you should instill it as soon as you remember about your missing. If it is the time for the next dose you should continue your regular dosing schedule. Do not instill a double dose to make up for a missed one.

Overdose

If an overdose occurs and you are not feeling well, stop taking the medication immediately. You should seek emergency medical attention or contact your healthcare provider immediately.

Storage

Store at cool room temperature, away from moisture and heat. Do not freeze. Keep the bottle tightly closed when not in use. Use within one month after opening a container. Keep out of the reach of children.

Side effects

The most common side effects associated with Gentalab® Eye and Ear Drops are:

  • irritation
  • itching
  • burning sensation
  • stinging sensation
  • inflammation of the skin (dermatitis)
  • sensitization or allergic reactions
  • hearing loss

Side effects occurrence does not only depend on the medication you are taking, but also on your overall health and other factors.

Contraindications

Do not use Gentalab® Eye and Ear Drops if you are allergic to Gentamicin (or to other Aminoglycosides), Chlorpheniramine, Zinc Sulphate.

Do not use Gentalab® Eye and Ear Drops if you are allergic to any of the ingredients in this medication.

Do not use Gentalab® Eye and Ear Drops if you have ever had severe allergic reactions associated with using antibiotics.

Do not use Gentalab® Eye and Ear Drops if you have myasthenia gravis, parkinsonism, reduced renal function.

Do not use Gentalab® Eye and Ear Drops in combination with such medications as capreomycin, cisplatin, loop diuretics (furosemide and etacrynic acid), teicoplanin, vancomycin, cephalosporin antibiotics, immunosuppressants (cyclosporin), amphotericin, capreomycin, colistin, polymixin, tacrolimus.

Do not use Gentalab® Eye and Ear Drops if you have ear infections with a perforated eardrum.

Do not use Gentalab® Eye and Ear Drops during pregnancy or lactation until considered absolutely necessary by your physician.

Do not use Gentalab® Eye and Ear Drops in infants below the age of 1 year.
